Over one million Americans and million people worldwide are infected with HIV making it one of the most pressing public health concerns of our time. Unfortunately there are no vaccines available and treatment requires lifelong anti retroviral therapies. This proposal combines two technologies for the development of a vaccine against HIVEnva novel adjuvant spontaneous nano liposome antigen particleization SNAP and a state of the art antigennative flexibly linked NFL trimers of HIV Enve gBG. This proposal will accomplish the following technical objectives Produce and provide well characterized cobalt porphyrin phosphoipid CoPoPCoPoP liposomes and NFL trimers Assess particleization parameters of His tagged BGNFL trimers with CoPoP liposomes Characterize conformational stability of particleized NFL trimers Determine the impact of antigen density in inducing HIV neutralization with rabbit SNAP immunization Assess multiplexed particleization parameters with His tagged BG and c Assess multivalent IgG response following rabbit SNAP immunization with BG and c and Characterize cobalt levels and His tag response in immunized subjects Rabbits will be vaccinated and sera tested for neutralizing antibodies to HIV The neutralizing titer of SNAP NFL vaccinated rabbits will be significantly higher than standard vaccination methods
