Many skin pathologies including photoaging, cancer, and inflammation may result from the effects of ultraviolet light on skin. Many of these pathologies are thought to be mediated by reactive oxygen species generated by light and subsequent inflammation. We will test the effects of topical antioxidants on these processes. We have learned how to stabilize ascorbic acid in solution and deliver it into skin in pharmacologic concentration (US patent #5,140,042). In this project we will test its ability to protect against ultraviolet induced photodamage as well as ultraviolet immunosuppression. The goal will be to develop claims for the product to assist in its commercialization. In addition, additives including vitamin E and sunscreens will be tested for their supplemental effect. Finally, novel ascorbic acid derivatives will be synthesized and tested for photoprotective activities in an attempt to develop more stable formulations for marketing.Proposed commercial application:Topical antioxidants may be useful in the treatment of skin aging, skin cancer, skin inflammation, skin photoprotection, wound healing, and skin purpura.National Institute of Arthritis and Muscoloskeletal and Skin Diseases (NIAMS)
