Enteric pathogen diagnostics for field use must be sensitive and fast, yet compact, robust, and facile with multiplex detection capability in complex matrices including feces. To address this challenge, NanoScience Diagnostics, Inc. (NSD) proposes to develop a set of immunomagnetic (antibody-magnetic bead) and antibody-quantum dot (QD) conjugate sandwich assays for the major enteric pathogens. This system will be able to capture, concentrate and purify enteric pathogens even in fecal matter by virtue of immunomagnetic separation. NSD has already demonstrated very sensitive antibody-QD fluorescence assays in neat buffers and concrete swabs (work done under the DOD contract No. W9132T-04-C-0030), which can now be enhanced by addition of antibody-magnetic bead concentration and purification of enteric pathogens. In Phase I, NSD will clearly demonstrate the feasibility of this approach in fecal matter for several pathogens or simulants. At the end of Phase II, NSD will deliver a working portable sensor and 200 prototype multiplexed assays to the Army for test and evaluation.

Benefits:
The combination of a multiplex magnetic bead-based antibody-quantum dot assay cartridge and handheld fluorescence assay reader would have powerful and lucrative applications in clinical point-of-care diagnostics, agricultural, environmental, food safety, and veterinary testing as well as homeland defense applications.
