| Aspen Aerogels Inc |
Northborough, MA |
Aspen Aerogel is a striking example of a firm coming to the event out of space that, at first blush, would appear to have remarkably little do with th-e primary areas of primary interest to Novartis OTC. Capitalizing on proprietary and patented advances in aerogel technology, the firm has developed a proprietary, easy-to-use blanket form of aerogels with broad application to a variety of markets including Oil & Gas, Building & Construction, Outdoor Gear & Apparel, Solar Thermal, Appliances and Transportation. Aspen Aerogels, Inc. provides aerogel technology for thermal/acoustic insulation, IR suppression, and shock absorption. AAI manufactures a unique flexible blanket (Cryogel, Spaceloft, and Pyrogel), powder, beads, and clear monolithic sheets. They are bringing nanotechnology to outdoor apparel, footwear, and other gear. Aspen's aerogel insulation, with its nano-sized air pockets that block cold and heat, offers higher thermal performance than any other insulation material - at a fraction of the thickness and weight. The firm has proven its aerogel insulation in boots, jackets, and sleeping pads tested in the most extreme environments on Earth - from the frigid summit of Mt. Everest to the blistering floor of Death Valley. A significant challenge in pharmaceutical development is achieving therapeutically acceptable solubility and bioavailability for poorly soluble drug compounds, while limiting toxicity. Aerogels are ultra low density, high surface area solid materials made by supercritical drying of wet gels. Literature has demonstrated that aerogel scaffolds can be loaded with small molecule drugs, whose dissolution profiles depend on the aerogel microstructures, and can range from immediate to prolonged release. There appear to be no commercial exploitation as yet; additionally, the patent landscape is sparsely populated. Aspen’s extensive experience designing, manipulating, processing, and testing aerogels positions the company to make efficient progress developing materials for this application. Academic relationships in this area are now in place and the firm plans to pursue government funding to corroborate our hypotheses. There would be significant value in a meaningful relationship with a company interested in commercial applications and willing to partner on the government funded R&D work. Aspen is developing aerogels as oral drug delivery systems to improve dissolution of BCS Class II and Class IV small-molecule pharmaceuticals. The firm has begun to test several OTC drug and aerogel composites, focused on immediate dissolution, to provide improved solubility and bioavailability and faster relief. By maximizing solubility, aerogel carriers allow immediate bioavailability which lowers the time taken for the drug to enter the bloodstream, total dosage required and drug toxicity. It is postulated that aerogel carriers increase drug solubility and dissolution due to its ability to stabilize the drug in an amorphous state, through interactions with the aerogel matrix. ...more |

| Nanocopoeia Inc |
St Paul, MN |
| Nanocopoeia is a drug delivery company developing a nanotechnology-enabled portfolio of fast-to-market dermal and transdermal therapeutic products. Th-e firm’s business model is to enhance the value and performance of off-patent and OTC, already proven drugs by creating novel nanoformulations optimized for delivery into and through the skin. Nanocopoeia has developed a method using the firm’s ENS process that consistently results in multiplefold improvements in water solubility, or more precisely, “nanoscale suspendability.” ENS is used to create either bare drug nanoparticles or coated, core:shell nanoparticles in a single, “bottom up” processing step rather than a multi-step “top down” process like wet milling or homogenization; both are techniques used to break down larger, insoluble particles; requiring secondary processing to create a monodisperse range of particle size. A further advantage of ENS produced drug formulations is the ability to produce coated nanoparticles in that single processing step. The firm proposes to develop nanoformulations of topically applied drugs for acne, alopecia, and nail fungus. The Nanocopoeia ElectroNanosprayTM technology is based on a particle production design that creates the nanoparticles from the bottom up. Starting with the base compound in a solution or suspension, nanoparticles are generated in an ultra-pure, very uniform state. The particles can also be coated in a single operation. The solvent evaporates during the production process and results in highly uniform nanoscale dispersions of drug. The competitive advantage of the Nanocopoeia enabling technologies resides in: • Particle purity and uniformity: the ability to generate sub-200 nm particles with tight control over particle size by controlling operating parameters • Single-step process for particle production: using a “bottom up” approach that operates at normal pressures and temperatures, thereby minimizing processing damage to active compounds • Coating capability: drug particles can be formed and coated in one step with an extremely broad range of polymers, excipients and surfactants • This same process can be used to create therapeutic nanoparticle coatings for medical devices. ...more |

Kansas City, KS |
| Orbis Bioscience provides novel controlled-release delivery systems to companies looking to reinvent their OTC products. Precision Particle Fabricatio-n produces uniform particles and capsules with narrow size distribution and exact control over particle structure. Clients use this elegant and robust solution to: 1) create differentiated products that are difficult for competitors to match, 2) innovate next-generation products with exactrelease profiles, and 3) leverage size uniformity for batch-to-batch reproducibility. Using this technology, Novartis could (1) gain unprecedented control of delivery rates for a variety of effects including fast action, rapid absorption, sustained release, and taste masking; (2) expand and improve delivery formats to include syrups, thin films, sprinkles, small pills, and sprays as alternatives to bulky tablets, patches, gums, or lozenges – offering flexibility in format options; and (3) improve drug performance with better exposure while reducing dosage frequency. Utilizing control of the particle and capsule physics, Novartis would have precise release rates and improved product quality through uniform, reproducible particles. Novartis would be able to achieve all of these improvements in a single step process by using Orbis’ PPF technology to manipulate key particle characteristics including size uniformity, shell thickness, and porosity. Orbis already works with multiple partners, leveraging the technology to enable new products through: (1) specific proof of concept development where Orbis develops customized formulations and the IP surrounding their manufacture, or (2) a research license in which Orbis provides the company with the technology for inhouse R&D and subsequent commercialization. Orbis’ exclusivity is on a product-by-product basis. There has been success with clients in the OTC space, and many other products still available for collaboration. ...more |
